I got it working after removing the quotes around MYSQLI_CLIENT_SSL.
Before:
define( 'MYSQL_CLIENT_FLAGS', 'MYSQLI_CLIENT_SSL' );
After:
define( 'MYSQL_CLIENT_FLAGS', MYSQLI_CLIENT_SSL );
