After a lot of searching around, it occurred to me that what I was trying to do was essentially to create a social network. Having search for WordPress Social Network plugins I discovered and am using Buddypress.

Solution was: RewriteBase /u Inside .htaccess
It is possible that: a) there are different values for the time set in .htaccess and wp-config. If it happens, WP uses the lower value. You should double check this. b) you run out of memory on your server. This could happen if you use shared hosting

As per the instructions for setting up WordPress MU Domain Mapping, sunrise.php needs to be placed directly in your /wp-content directory.
